Solid tissue culture application in cytogenetic analysis

Abstract
Background: In human diagnostics, chromosomal analysis of solid tissue cultures is essential for confirming genetic suspicion diagnoses via cytogenetic or molecular techniques. Cytogenetic analysis of solid tissues is the preferred method when chromosomal studies of other tissues fail to yield a definitive diagnosis. Methods: A retrospective, cross-sectional study was conducted using databases from the cytogenetics laboratories of the National Center of Medical Genetics in Havana, Cuba, and the National Institute of Medical Genetics and Genomics in Panama City, Panama. The cohort included 67 solid tissue cultures: 38 derived from skin biopsies, 18 from products of conception/aborted fetuses, nine from gonadal tissues, and two from tumors. The analysis was carried out using banding cytogenetics with a resolution of 450–550 bands. Results: The most frequent clinical indication was disorders of sex development, followed by studies on fetal tissue after an abortion. Chromosomal aberrations were identified in 22% of the analyzed tissues, with chromosomal mosaicism for aneuploidies being the most prevalent form. Two cases of polyploidy and three structural aberrations were identified. Furthermore, two cases of Swyer syndrome were detected. Conclusions: Solid tissue culture may enable accurate cytogenetic diagnosis in cases where chromosomal aberrations are tissue-specific. Additionally, it is a reliable method for identifying aberrant cell lines in cases of low-level chromosomal mosaicism.
